Kinetics of T-cell Killing in Three Dimensional Tumor Spheroids



                3-D tumor spheroids more closely resemble the tumor microenvironment than traditional 2-D cell cultures. Past cancer models have focused on the interactions of effectors and targets at either the 2-D or 3-D level, but not both. Understanding the differences between effector cell and tumor cell interactions in 2-D versus 3-D will help shed light on the mechanisms that make tumor spheroids more resilient. Here, we employed TALL-104 cells and DAOY cells as an ideal model to study the kinetics of cancer cells and effector cells at both the single cell level and at the 3-D. A Timing assay was run to gather information about rate constants and in conjunction, the same set of TALL-104 cells was added to DAOY spheroids and studied using the Incucyte system. Two different 3-D models were created and fitted to the experimental data. The rate constants were then compared between the 2-D and 3-D models.
